熱帶疾病醫療暨防治中心長期研究熱帶疾病,特別是登革熱及其他蚊媒傳染病,與國內、外學者及廠商合作,包含夏威夷大學王維恭教授、新加坡國立大學Justin Chu Jang Hann副教授、國家衛生研究院陳俊宏老師、成功大學彭貴春教授等。

 

  • 登革熱臨床表現與基礎研究:與台南國立成功大學的彭貴春教授合作對急性感染期的病毒和宿主免疫反應進行了分析。藉由探討登革熱病毒感染期間造血幹細胞的活動,證明登革熱病毒感染可以誘導造血幹/前驅細胞(Hematopoietic stem and progenitor cell)活化、此活化的細胞不僅可被登革熱病毒感染,而且與病毒一同培養後可使登革熱病毒恢復感染性。這是嶄新的發現,對登革熱病毒如何入侵人類、傳播和發病機制提供了另一個探討對象。

  • 檢驗方法的驗證與比較:我們驗證多種RT-PCR 系統和快速診斷測試,以檢測台灣南部、索羅門群島發燒患者的急性樣本和蚊子中的登革病毒感染並與國家衛生研究院團隊合作檢測蚊子中的登革病毒,同時比較快篩試劑性能。

  • 血清學調查及檢測方法研發:與夏威夷大學王維恭教授合作,從研究登革病毒感染後的多克隆抗體和單克隆抗體開始,開發不同的血清學測試來區分登革病毒、茲卡病毒和其他黃病毒。這些包括以NS1為基礎的ELISA、MIA及以VLP為基礎的ELISA和西方墨點法分析。同時也比較登革病毒不同檢測原理的ELISA方法在血清陽性率研究中的性能,並調查2015年登革熱大爆發之後,高雄的登革熱病毒血清陽性率。與日本早稻田大學伊藤悅朗副教授合作,開發超高靈敏度酵素免疫分析法(Ultrasensitive ELISA with Thio-NAD Cycling),性能媲美核酸檢測方法,並且更低成本、易於使用,適合低收入國家。

  • 登革熱疫苗及登革熱重症、治療及登革病毒基因相關研究:今年發表一篇合作論文,針對Dengvaxia登革熱疫苗在菲律賓兒童施打後,突破性感染風險分析,同時驗證上述新開發ELISA方法。研究發現接受Dengvaxia登革熱疫苗接種的登革病毒血清陰性兒童在突破性登革病毒感染後出現嚴重疾病,使得接種疫苗後的兒童得到嚴重登革熱的風險更高。開發並驗證一種能夠識別DENV-naïve Dengvaxia登革熱疫苗接受者的檢測方法至關重要。這將用於對突破性登革熱病毒感染進行更頻繁和更廣泛的評估並及時治療。
